DNA MAPPING

Genome mapping is the creation of a genetic map assigning DNA fragments to chromosomes.

When a genome is first investigated, this map is nonexistent. The map improves with the scientific progress and is perfect when the genomic DNA sequencing of the species has been completed. During this process, and for the investigation of differences in strain, the fragments are identified by small tags. These may be genetic markers (PCR products) or the unique sequence-dependent pattern of DNA-cutting enzymes. The ordering is derived from genetic observations (recombinant frequency) for these markers or in the second case from a computational integration of the fingerprinting data. The term "mapping" is used in two different but related contexts.

Two different ways of mapping are distinguished. Genetic mapping uses classical genetic techniques (e.g. pedigree analysis or breeding experiments) to determine sequence features within a genome. Using modern molecular biology techniques for the same purpose is usually referred to as physical mapping.

RNA

Ribonucleic acid (RNA) is a biologically important type of molecule that consists of a long chain of nucleotide units. Each nucleotide consists of anitrogenous base, a ribose sugar, and a phosphate. RNA is very similar to DNA, but differs in a few important structural details: in the cell, RNA is usually single-stranded, while DNA is usually double-stranded; RNA nucleotides contain ribose while DNA contains deoxyribose (a type of ribose that lacks one oxygen atom); and RNA has the base uracil rather than thymine that is present in DNA.

RNA is transcribed from DNA by enzymes called RNA polymerases and is generally further processed by other enzymes. RNA is central to protein synthesis. Here, a type of RNA called messenger RNA carries information from DNA to structures called ribosomes. These ribosomes are made from proteins and ribosomal RNAs, which come together to form a molecular machine that can read messenger RNAs and translate the information they carry into proteins. There are many RNAs with other roles – in particular regulating which genes are expressed, but also as the genomes of most viruses.

Genetic lung cancer is higher in women, nonsmokers and individuals with early onset the cancer. Taken as a whole, it has been predicted that 1.7% of cancers of lung up to the age of 68 are hereditary.

Lung cancer is the third most common type of cancer identified in the United States and is the primary cause of death in the United States and in the world. While it is recognized that smoking leads to cancer of lung, there are individual differences in vulnerability to lung carcinogens.

Smokers who develop lung cancer are less probable to suffer a family history than nonsmokers that develop the cancer. However, that said, for individuals who have a genetic predisposition to the cancer, smoking comes out to strengthen that risk.

However, Genetics also play a significant role in the development of this deadly disorder. It's likely for heavy smokers to stay cancer free during their lives, while non-smokers can still develop lung cancer because of their genetic makeup.

If you realize that you're genetically predisposed to cancer of lung, the most effectual measure of prevention is not smoking cigarettes. Other measures to stay healthy also could reduce your risk. The variety of leopard gecko patterns (known as morphs - that is, types that are visually distinct from each other because of color and pattern variations) available today is truly staggering. If you've only ever seen the standard 'wild' leopard gecko, then you owe it to yourself to check out some pictures of spectacular morphs such as the Diablo Blanco, Super Hypo Carrot Tail, or Rainwater Mack Snow. They look as amazing as their names suggest.

Seeing these leo morphs might well get you interested in breeding leopard geckos to produce them, and why not - it's a great hobby. However, the sting in the tail is that doing this requires you to become an amateur geneticist, as otherwise you simply won't be able to understand well enough how to get the results you want.

The first thing to learn is some frequently used terms. Phenotype, for example, refers to the traits that a creature has and is showing. Genotype on the other hand refers to the traits that a creature is carrying but not necessarily showing. A couple more terms: heterozygous (or just 'het' for short) refers to a creature that is carrying one copy of a specific recessive trait and one copy of a normal trait. Homozygous is the opposite: it refers to a creature that is carrying two copies of the same trait.

Ok, let's now use those terms: a leopard gecko of the albino phenotype is homozygous. It looks like an albino because it carries two copies of the recessive albino trait. A het albino leo, on the other hand, won't look like an albino, as it carries only one copy of the recessive albino trait. It's important to know when you have a leo of the het albino phenotype however, as otherwise you might just think it's a normal wild-type leopard gecko, and miss the opportunity to breed it with another het albino to try to produce a homozygous albino.

If you have two het albinos - a male and a female - then each of their children has a 25% chance of being a homozygous albino, and therefore looking like an albino. If you have two albinos, then all of their children will be of the albino phenotype too. Simple enough?

The other important concept to grasp is that of 'line bred' traits. Line breeding refers to selecting leos with a certain trait that's a little different from the norm and breeding them together over successive generations to emphasize that trait. The tangerine morph, for example, was developed by picking a male and a female leopard gecko that both happened to have a slightly brighter background color than usual and breeding them together, then picking the brightest of their children and breeding them together, and so on and so on over a number of generations. Over time a really bright tangerine morph was produced.

Genetic engineering today is no longer a new term for the world. Every day in the newspapers, televisions, magazines the new inventions of genetic engineering are noticed. Genetic engineering may be described as the practice that manipulates organism's genes in order to produce a desired outcome. Other techniques that fall under this category are: recombinant DNA technology, genetic modification (GM) and gene splicing.

The roots of genetic engineering are connected to the ancient times. The Bible also throws some light on genetic engineering where selective breeding has been mentioned. Modern genetic engineering began in 1973 when Herbert Boyer and Stanley Cohen used enzymes to cut a bacteria plasmid and inserted another strand of DNA in the gap created. Both bits of DNA were taken from the same type of bacteria. This step became the milestone in the history of genetic engineering. Recently in 1990, a young child with an extremely poor immune system received genetic therapy in which some of her white blood cells were genetically manipulated and re-introduced into her bloodstream so that her immune system may work properly.


Genetic engineers hope that with enough knowledge and experimentation, it will be possible in the future to create "made-to-order" organisms. This will lead to new innovations, possibly including custom bacteria to clean up chemical spills, or fruit trees that bear different kinds of fruit in different seasons. In this way new type of organisms as well as plants can be developed.

Genetic engineering requires three elements: the gene to be transferred, a host cell into which the gene is inserted, and a vector to bring about the transfer. First of all, the necessary genes to be manipulated have to be 'isolated' from the main DNA helix. Then, the genes are 'inserted' into a transfer medium such as the plasmid. Third, the transfer medium (i.e., plasmid) is inserted into the organism intended to be modified. Next step is the element transformation whereby several different methods including DNA guns, bacterial transformation, and viral insertion can be used to apply the transfer medium to the new organism. Finally, a stage of separation occurs, where the genetically modified organism (GMO) is isolated from other organisms which have not been successfully modified.
